Why Do ChIP-seq and RNA-seq Integration Analysis?
ChIP-Seq specifically enriches target protein A-bound DNA fragments by chromatin immunoprecipitation (ChIP) technology and sequences them. The millions of sequence tags obtained are precisely localized to the genome by bioinformatics analysis, thus obtaining genome-wide information on DNA segments that interact with histones, transcription factors, etc. In addition, RNA-seq was performed to find the downstream genes affected by gene A by overexpression or knockdown / disruption of gene A. Later, the target genes and binding sites of target protein regulation were screened by real time PCR and double luciferase system and other experimental validation.
