Why Do RNC-seq and RNA-seq Integration Analysis?

Much of the purpose of studying the translatome is to investigate the portion of the sample genes that are being translated to synthesize proteins and the translational changes in the genes under different experimental treatment conditions. Only in combination with the transcriptome can post-transcriptional regulation be studied in more depth. The Ribosome Nascent-chain Complex (RNC) is the most important unit in the translation process. the ratio of RNC-mRNA to total mRNA is the translation ratio (TR). The TR is related to the phenotype of the cell and is also regulated by UTR, miRNA, etc. Comparing TR-altered genes in different samples can study the molecular mechanisms of disease development, drug action, development, etc. It also provides a powerful tool to elucidate the molecular mechanisms of ncRNAs such as miRNAs.

Probing the mechanism of a gene's effect on a disease

(1) Screening genes that are in high translational efficiency among genes being translated and exploring their global functional impact on disease to guide subsequent experiments.

(2) Exploring the impact of unconventional translation phenomena on disease.

Deep transcriptome sequencing analysis

(1) Identify the genes being translated in differentially expressed genes and the efficiency of translation.

(2) Narrowing the range of differentially expressed genes in the transcriptome, screening for genes that are being translated, or comparing their translation efficiencies.

(3) Expand the range of differentially expressed genes in the transcriptome, then additional differentially expressed / translated genes can be identified through transcriptomic analysis.

Explain the discordance between transcriptomic and proteomic results

If transcriptomics shows differential or significant differences in gene expression, while proteomic validation reveals no differences or insignificant differences, possibly because the genes are transcribed but not translated or translated inefficiently.

Probe whether a certain lncRNA is translated into a short peptide or protein.
