Why Do Integrative Analysis of RNA-seq and 16S?

Why Do Integrative Analysis of RNA-seq and 16S

In the study of the association mechanism of microorganisms and hosts, 16S analysis obtained information such as species distribution characteristics of samples and different species between groups, while transcriptomics can reflect the expression activity of hosts. In order to further explore the specific molecular mechanism of the relationship between the health, disease, growth, and stress resistance of microorganisms and hosts (humans, animals and plants, etc.), the association analysis of species and genes / functions can also be carried out. Therefore, in order to identify which species are associated with which gene expression from the huge collection of microorganisms and genes, and the differences in grouping arise, it is necessary to perform 16S-transcriptomics association analysis.

Bioinformatics Analysis

Based on the sample data of one-to-one correspondence between microorganisms and transcriptomes, in the integrated analysis, we will use three models: (1) O2PLS model (bidirectional orthogonal projections to latent structures), which uses data on species abundance and gene expression to build a model. Through model prediction, the associated species and gene sets are obtained. (2) Correlation coefficient model (sample group ≥ 3), calculate the pearson correlation coefficient of species abundance and gene expression, and display them in heat map and network diagram. (3) CCA model (canonical correlation analysis), which analyzes the relationship between genes and species distribution in the form of dimensionality reduction (the gene name used for CCA analysis needs to be provided).
