Why Do Ribo-seq and Whole Transcriptomics Integration Analysis?

The process of translating RNA into protein is not simply the transfer of genetic information, but the precise translation and regulation to produce a protein of a specific sequence and structure.

Ribosome Profifiling (or Ribo-seq) is the process of enriching, sequencing and quantifying ribosome-bound mRNA fragments (approximately 30 nt in length) that are being translated. This allows us to obtain the distribution of ribosomes on each transcript, to infer the position of initiation codons (including non-ATG initiation) and uORFs, to analyze the efficiency of translation, and to resolve the mechanisms of translation regulation.

Integration of the translatome with the whole transcriptome can reveal the translation efficiency of mRNA and the reliable small open reading frames (sORFs) of non-coding RNA. In this way, we can systematically analyze the association mechanisms between translation group and whole transcriptome data, such as reduced translation efficiency and unchanged transcript levels leading to decreased protein levels; as well as identify reliable sORFs in non-coding RNAs (especially lncRNA) to provide guidance directions for predicting their encoded small peptides.
